A Study of Potassium Permanganate ' Fixation ' for Electron Microscopy
نویسنده
چکیده
The action of buffered potassium permanganate as a fixative for electron microscopy has been investigated. Electron contrast has been shown to be produced by the deposition of granular material in the tissue. The particles are about 50 A in diameter. Actual fixation of the tissue is performed by the dehydrating alcohol. Histochemical studies have shown that RNA and histones are removed, whereas phospholipid-protein complexes are 'unmasked'. The reaction of the permanganate with unmasked protein gives rise to high membrane contrast in electron micrographs.

Permanganate—A New Fixative for Electron Microscopy
During the early phases of tissue sectioning for electron microscopy, virtually all of the standard cytological fixatives were tried. Of these, osmium tetroxide appeared to give the best preservation, and this was further improved by buffering the fixative solution at a slightly alkaline pH (Palade (1)). A variant developed by Dalton (2) contains Os04 in a chromate-dichromate system. Formalin h...

A comparative study of the fixation of isolated rat liver mitochondria was undertaken. If the criterion is adopted that after processing, the mitochondria should resemble as closely as possible rat liver mitochondria in situ, the procedure found to produce such preservation was that of fixation in suspension in veronal-buffered 2% potassium permanganate. Fixation in osmium tetroxide produced va...
متن کاملElectron Microscope Studies of the Human Epidermis The Clear Cell of Masson (Dendritic Cell or Melanocyte)
The human epidermis has been studied by electron microscopy following osmium tetroxide and potassium permanganate fixation. An anatomically distinct cell in the human epidermis has been demonstrated with features similar to the melanocyte of the hair bulb described by Barnicot, Birbeck and Cuckow (3). It is dendritic in form and does not contain tonofilaments. "Intercellular bridges" are not fo...
متن کاملSodium Permanganate Fixation for Electron Microscopy
Heightened interest in the cytology of certain free-living flatworms has been evident in the literature throughout the past year (5, 6, 10-12). The variety of problems dealt with by various investigators reflects the abundance of curious cellular specialization found in this group of animals. Moreover, the suitability of the Turbellarians for the study of regenerative processes is well recogniz...
متن کاملThe development of the nervous system in chick embryos, studied by electron microscopy.
Tissue from 73 chick embryos incubated for periods between 1 and 13 days has been studied. Before fixation each specimen was staged according to the normal table of Hamburger & Hamilton (1951). The tissue was fixed either in osmium tetroxide, or in potassium permanganate (Luft, 1956). In either case the pH was buffered to 7-4 with sodium veronal (Palade, 1952) and fixation was at 0° C. In the e...
